Entacapone is an anti-Parkinsonian drug, successfully developed by OrionPharma of Sweden, as a highly selective and potent As of levodopa, dopamine and DOPAC and significantly reduces the dose required to increase striatal dopamine concentrations. The combination of levodopa and carbidopa with COMTI significantly increased the bioavailability of levodopa (3- to 4-fold). The activity of COMTI in erythrocytes is reversible, with a maximum inhibitory activity of 82% at a dose of 800 mg, so entacapone in combination with levodopa and carbidopa can be used as an adjunctive therapy in primary Parkinson's disease.
