Mecillin is an amphoteric compound with the molecular formula C15H23N3O3S. It is a white or off-white powder, very soluble in water and the aqueous solution is clear, colourless to yellow. It was synthesized by FrantzJLund and J. Tybring of Leo Pharmaceuticals in Denmark in the early seventies. It is a third generation broad-spectrum semi-synthetic penicillin, a new compound found in the semi-synthetic amidine-based semi-synthetic penicillins. It differs from other penicillins in that the C6 position of the parent nucleus of the penicillin is replaced by a squint group in place of the usual phthalamide bond, thus conferring unique antibacterial activity. It acts mainly on bacterial transpeptidases. It is particularly potent against Gram-negative bacteria, especially Escherichia coli, S. pneumoniae, Enterobacter spp, Acinetobacter cloacae, Bacillus dysenteriae and Salmonella. Methicillin is the most potent of the squint-based penicillins. It is poorly absorbed orally, but can be given intramuscularly or intravenously.
