[Source Plant] Lentinus edodes is the fruiting body of the fungus Lentinus edodes of the genus Lentinus edodes. Lentinus edodes is rich in nutrition. Every 100 grams of dried product contains 13 grams of protein, 1.8 grams of fat, 54 grams of carbohydrates, 7.8 grams of crude fiber, and a variety of vitamins. It contains more vitamin D and adenine, which has a preventive effect on the occurrence of rickets and liver cirrhosis. Lentinus edodes contains lentinan, which can inhibit the proliferation of sarcoma S-180 in mice and has certain prospects in the development of natural anticancer drugs. The artificial cultivation of shiitake mushrooms began in China. Wang Zhen's "Nongshu" in Yuan Dynasty recorded the cultivation method of shiitake mushrooms in detail. This natural spore inoculation and cultivation method has been used in China, Japan and other countries until modern times. Polysaccharides | Any sugar that can be hydrolyzed into multiple monosaccharide molecules or their derivatives is a polysaccharide. It is a macromolecular compound formed by condensation and dehydration of 20 monosaccharides to tens of thousands of monosaccharides. research history | In 1969, Japan's Chihara Kuro first used hot water to extract 6 kinds of extracellular polysaccharides from the fruiting body of Lentinus edodes, of which 2 have obvious anti-tumor activity, with an average relative molecular mass of 1000000, belonging to branched β(1 → 3) glucan. in 1970, Chihard and others extracted 4 kinds of polysaccharides from shiitake mushrooms by hot water extraction and organic solvent precipitation, of which 2 kinds of polysaccharides have anti-tumor effect. in the early 70s, Shida et al. extracted three water-soluble polysaccharides from Lentinus edodes by trichloroacetic acid extraction, methanol precipitation and other methods, one of which was composed of Gal,L-Fuc and Man (molar ratio 6:1.5:1). this is another anti-tumor substance reported for the first time in Lentinus edodes except β-glucan. Sasaki also reported that besides the macromolecule lentinan with a relative molecular mass of 1 million, another small molecule lentinan with a relative molecular mass of 16000 also has anti-tumor activity. Since the 1980s, the research on lentinan has been more in-depth. For example, Schulty extract an intracellular polysaccharide from the hyphae of lentinus edodes, the relative molecular mass (6.0~9.5)× 104 is mannoside containing α-chain, accompanied by a small amount of peptide immunoactive substances composed of serine, threonine and alanine. In 1980, Cotoneaster II in Japan confirmed that lentinan was a broad-spectrum immune promoter. A large number of subsequent studies have shown that lentinan has anti-tumor effects, especially for chronic myeloid leukemia, gastric cancer, nasopharyngeal cancer, rectal cancer and breast cancer. Lentinan has a triple helix three-dimensional structure. When this three-dimensional structure is destroyed by urea and DMSO, the immune activity disappears. Other polysaccharides in Lentinus edodes are composed of glucose, galactose, mannose, L-arabinose and xylose, and there are also α-mannosides derived from the mycelium of submerged fermented Lentinus edodes. In recent years, people have carried out research on chemical modification of lentinan to find more effective polysaccharide substances. For example, 10 μg/mL sulfated lentinan can effectively protect HIV-infected MT-4 cells, and 100 μg/mL can inhibit RT activity and the generation of synaptic cells. Its inhibitory effect on stress transcriptase is higher than that of sulfated dextran. Through 13C-NMR and other measurements, it is proved that the introduction of sulfuric acid base leads to the physical and chemical properties of lentinan and the conformational changes of the main body, resulting in the different immune activity of the derivatives. |

Then remove the mushroom residue, the supernatant is dialyzed or ion exchange to remove small molecular impurities, and the crude product is obtained after precipitation with ethanol or thiamine. All precipitates were collected by centrifugation and washed with ethanol. It was suspended in 1.2L 20% acetic acid and stirred at 0 ℃ for 5min. The precipitate was collected by filtration, and 1L 50% acetic acid was stirred at 0 ℃ for 3min. The precipitate was collected by centrifugal separation and then dissolved in 2L 6% sodium hydroxide aqueous solution. After centrifugation to remove impurities, add 4L ethanol. Collect the precipitate, wash it twice with ethanol, then wash it once with ether, and dry it in vacuum to obtain powder.
