Product Description:

D-amino acid oxidase (DAAO, DAO, EC 1.4.3) is a typical flavin protease with FLAVin adenine (FAD) as a cogroup. It oxidizes the amino group of D-amino acids to produce the corresponding ketone acid .  D-amino acid oxidase has high stereoisomerism selectivity and broad spectrum for catalytic reaction substrates, which can be widely used for qualitative and quantitative analysis of D-amino acids, biosensors, production of L-amino acids and A-ketoic acids.

Definition of activity: One unit will oxidatively deaminate 1.0 μ mole of D-alanine to pyruvate per minute at pH 8.3 at 25 °C, in the presence of a catalase.

D-amino acid oxidase binds to GL-7-ACA acylase, which plays a major role in the conversion of CPC to 7-ACA by two-step enzymatic method.

(1) L-amino acid oxidase, which exists in kidney and liver mold, snake venom and bacteria, is the aerobic dehydrogenase that directly oxidizes L-amino acids. EC1.4.3.2. If catalase is not present, H2O2 is decarboxylated to form ketoic acid.

Its coenzyme is FMN or FAD. The substrate has a wide specificity and can oxidize leucine and methionine well, but does not act on aspartic acid, glutamic acid and glycine. However, the enzymes in bird liver are specific to basic amino acids. However, because of their low activity, it is questionable whether animal enzymes are meaningful for amino acid metabolism. However, the oxidation activity of L-α -hydroxyl acid is strong, which may have this significance. Snake venom has an enzyme of about 130,000 molecular weight and contains 2 molecules of FAD.

(2) D-amino acid oxidase. An enzyme that produces ketoacids for the oxidation of D-amino acids. EC1.4.3.3.

D-amino acid +O2→ ketoic acid +H2O2+NH3 with FAD as coenzyme, widely exists in organisms. It is also common in kidney and liver of animals, but its significance is not clear. Because it reacts well with D-α -carboxylic acids, it may also be a true substrate. It acts best on d-proline, but also on many other amino acids. The crystallase of porcine kidney had a molecular weight of 115,000 and contained 2 molecules of FAD.
